Wouldn't have thought so.
Pump has 2 non-return valves in it, so the diaphragm bounces back and forth, pulling the fuel through 1 valve, and pushing it through the other.
(I would think) without actual fuel in it, the no return valve seals would not be strong enough to give you suction.
To test, what I have done is pull the fuel hose off the carb entry, and put in a jar.
When you put the ignition on, you should have a steady, strong flow of fuel into the jar.
